Category Name Pain Killers / Systemic Enzyme & NSAID Combination / Anti-Inflammatory & Analgesic
Common Indications TRYVAIN-BR is indicated for the relief of pain, inflammation, and swelling associated with traumatic, surgical, degenerative, and inflammatory musculoskeletal conditions. It is commonly prescribed for:
- Arthritis-related pain including rheumatoid arthritis and osteoarthritis
- Post-operative pain, swelling, and tissue oedema following surgical procedures
- Sports injuries, sprains, and strains with associated inflammation and bruising
- Bone fractures with associated soft tissue swelling and pain
- Post-traumatic inflammation and oedema requiring combined enzymatic and anti-inflammatory therapy
- Degenerative joint and muscle conditions requiring long-term anti-inflammatory support
- Soft tissue injuries with impaired circulation and localised swelling
- Tendinitis, bursitis, and periarticular inflammatory conditions
- Dental and oro-facial post-operative pain and swelling
Common Dosings One tablet orally, twice to three times daily — preferably on an empty stomach for optimal enzyme absorption — or as directed by your physician. The dose and duration of therapy should be determined by the treating physician based on the condition being treated, severity of symptoms, and clinical response. Use the lowest effective dose for the shortest duration necessary to control symptoms. Do not exceed the prescribed dose or use for prolonged periods without medical supervision.
Common Side Effects No commonly reported side effects have been documented for this specific combination at recommended therapeutic doses. However, based on the individual components, the following may occasionally occur. Consult your doctor if any of these persist or worsen:
- Nausea or mild gastrointestinal discomfort
- Abdominal bloating or flatulence
- Skin rash or allergic reaction — inform your doctor promptly
- Signs of gastrointestinal irritation such as stomach pain or heartburn — associated with the Diclofenac component
- Signs of gastrointestinal bleeding such as black or tarry stools or vomiting blood — seek immediate medical attention if these occur
- Serious allergic reactions such as facial swelling, difficulty breathing, or severe skin reactions — seek immediate medical attention if severe
Common Interactions Inform your doctor about all medications, supplements, and herbal products you are currently taking. Notable interactions include:
- Anticoagulants (Warfarin) / Antiplatelet Drugs (Aspirin, Clopidogrel): Bromelain's fibrinolytic properties combined with the anti-inflammatory effects of Diclofenac may significantly increase the risk of bleeding complications when used concurrently — regular INR monitoring and close clinical observation are strongly recommended; avoid combination use where possible
- Antibiotics (Amoxicillin, Tetracycline): Bromelain may enhance the intestinal absorption and plasma levels of certain antibiotics, potentially increasing both their therapeutic effects and the risk of antibiotic-related side effects — inform your doctor if concurrent antibiotic therapy is prescribed
- Other NSAIDs / Corticosteroids: Concurrent use with Diclofenac may increase the risk of gastrointestinal irritation, ulcers, and bleeding — avoid combination use without strict medical supervision
- Anti-Inflammatory Supplements (Fish Oil, Garlic, Ginger, Ginkgo): May add to the anticoagulant and fibrinolytic effects of Bromelain and Rutoside, increasing the overall risk of bleeding — use with caution and inform your doctor of all supplements being taken
- ACE Inhibitors, ARBs & Diuretics: Diclofenac may reduce the effectiveness of antihypertensive and diuretic medications and increase the risk of renal impairment — use with caution under close medical supervision
- Lithium & Methotrexate: Diclofenac may increase the plasma concentrations of these drugs, raising the risk of serious toxicity — concurrent use requires careful clinical and laboratory monitoring
Special Instructions
| Condition | Guidance |
|---|---|
| Pregnancy | Adequate safety data during pregnancy is not currently available for this combination. Diclofenac is particularly contraindicated during the third trimester. Use only under explicit medical direction after careful benefit-risk evaluation by the treating physician. |
| Breastfeeding | Adequate safety data during breastfeeding is not currently available. Consult your doctor before use and consider a safer alternative during lactation. |
| Alcohol | The safety of concurrent alcohol use has not been fully established. Alcohol may increase the risk of gastrointestinal bleeding, gastric ulceration, and liver stress — it is strongly advisable to avoid alcohol during therapy. |
| Driving & Machinery | May affect driving ability in some patients — particularly if associated electrolyte or metabolic fluctuations occur. Exercise caution and avoid driving or operating heavy machinery if you experience dizziness, visual disturbances, or impaired concentration during therapy. |
| Liver Disease | Unsafe to use in patients with liver disease and should be avoided. Diclofenac is extensively metabolised in the liver — use is contraindicated in significant hepatic impairment. Consult your physician immediately if liver disease is diagnosed during therapy. |
| Kidney Disease | Unsafe to use in patients with kidney disease and should be avoided. Diclofenac may further compromise renal blood flow and kidney function — use is contraindicated in significant renal impairment. Consult your physician before use. |
Mechanism of Action TRYVAIN-BR combines four active ingredients, each working through a distinct and complementary mechanism to provide comprehensive pain relief, anti-inflammatory action, and accelerated tissue healing:
Trypsin (48 mg): A serine protease enzyme that exerts its therapeutic effect by breaking down abnormal proteins, fibrin deposits, and inflammatory exudates at the site of tissue injury. This proteolytic action reduces localised oedema, improves microcirculation and lymphatic drainage at the injury site, enhances the removal of damaged cellular debris, and accelerates the natural tissue repair process. Trypsin also stimulates the production of endogenous anti-inflammatory mediators, further reducing pain and swelling
Bromelain (90 mg): A potent proteolytic enzyme complex derived from pineapple stem that works by selectively degrading fibrin and inflammatory proteins at the site of injury or post-operative swelling. Bromelain reduces prostaglandin-mediated inflammation, inhibits platelet aggregation, enhances microvascular circulation, and reduces tissue oedema and bruising. Its fibrinolytic activity helps resolve post-traumatic and post-surgical swelling more rapidly than conventional therapy alone
Rutoside Trihydrate (100 mg): A bioflavonoid and potent antioxidant derived from Rutin that works by neutralising reactive oxygen species (free radicals) generated at the site of inflammation and tissue damage. By protecting endothelial cells from oxidative injury, Rutoside reduces capillary permeability and fragility, decreases fluid leakage into surrounding tissues, and helps resolve oedema. Its antioxidant and anti-inflammatory properties complement and enhance the enzymatic actions of Trypsin and Bromelain
Diclofenac Sodium: A potent non-steroidal anti-inflammatory drug (NSAID) that exerts its therapeutic effect by non-selectively inhibiting cyclo-oxygenase (COX-1 and COX-2) enzymes, thereby reducing the biosynthesis of prostaglandins — the key chemical mediators responsible for triggering and sustaining pain, inflammation, fever, and swelling at the affected site. Its rapid and potent peripheral analgesic and anti-inflammatory action provides immediate symptomatic relief while the enzymatic components simultaneously address the underlying tissue oedema and promote healing
Together, the systemic enzymatic action of Trypsin and Bromelain, the antioxidant and capillary-protective effect of Rutoside, and the potent anti-inflammatory and analgesic action of Diclofenac provide a comprehensive multi-mechanism approach to pain and inflammation management — delivering faster, more complete, and more sustained relief than conventional single-agent NSAID therapy alone.
Instructions to Take
- Take this tablet exactly as prescribed by your doctor
- For optimal enzyme absorption, take on an empty stomach — at least 30 to 60 minutes before meals or 2 hours after meals — unless stomach discomfort occurs, in which case take with a light meal and consult your doctor
- Swallow the tablet whole with a full glass of water — do not crush, split, or chew
- Take at regular intervals as prescribed to maintain consistent drug levels and sustained pain relief
- Use the lowest effective dose for the shortest duration required to control symptoms
- Do not combine with other NSAIDs, aspirin, or anticoagulant medications without medical advice
- Inform your doctor of all supplements being taken — particularly those with anticoagulant or anti-inflammatory properties
- Do not self-medicate, adjust the dose, or continue therapy beyond the recommended duration without consulting your doctor
- If a dose is missed, take it as soon as you remember — however, skip the missed dose if it is almost time for the next scheduled dose. Do not double the dose to make up for a missed one
- Avoid alcohol throughout the course of treatment
- Maintain adequate hydration during therapy to support renal function and tissue circulation
- Keep out of reach of children
- Store in a cool, dry place away from direct sunlight and moisture
